Security software was my first thought but if it's happening on a Mac and a PC that seems unlikely unless you're running the same product on both. Is there any filtering software on your home router? For your ISP to block just you from just one site would be pretty bizarre. Apart from rebooting all your network hardware, I'm afraid I'm out of ideas

I called my ISP and asked them if it was possible that they would have blocked this site. They said, no, they cannot do that. They can shut down my access completely, but they do not have the ability to block access to one particular site. This is reassuring. The next thing I'm going to do is take a laptop from my work-place home with me and see if I can connect using that. This is a laptop that has just been wiped clean and a fresh copy of WinXP (with updates) installed. I've checked it here at work to confirm that I can access the site from work. Later today I'll test to see whether I can access it from home using the laptop from work.... stay tuned.

I won't put much more time into this effort, but just wanted to relate that the laptop I brought home from work last evening would not connect to mp3sugar either. It would connect from work, but once I brought it home and tried to connect there, it would not. I also rebooted my cable modem and router, to no avail. If I get a chance, I'm going to ask some others around my workplace whether they are able to connect to the site from their homes. Why anyone would single this site out to block while letting the other Russian music sites be accessible is beyond me. I am completely stumped.

Have you tried checking in with Mp3sugar techsupport and have them
do a check on your ISP address and make sure there is nothing wrong
from their side.Sometimes you can be blocked in error by their software.
Have them check it.

If you're only blocked at DNS-level (and it seems to be that way, since you are able to use Tor to circumvent the blocking), then I would suggest you to use this excellent step by step guide made by solaris:

I came home from work this evening, and thought I would try to access mp3sugar one more time. Lo and behold, up it came!!

Now I'm thinking there might have been a router down somewhere upstream, or perhaps a router's interface went down temporarily somewhere.

You ought to follow the advice of viracocha, though - and use OpenDNS instead of your ISP's ditto
